Introduction:
HIV is a family of retrovirus carrying RNA as its genome. Previously it was known as TLV (T lymphocyte virus) as it infects T helper cell and reduces CD4 receptor to cause immunodeficiency in human body. At present the HIV infection rate is very high all over the world and in India this is pretty much alarming. Though there has no drugs been found which could destroy HIV genome due to its high mutation frequency, Zidovudine is used to control as it prevents reverse transcriptase to synthesize cDNA which is later integrated in the host genome. But any time zidovudine can lose its efficiency as mutation at HIV genome (codon 67, 70,215,219) will change RT property as it is site specific and mutation cause it lose its specificity.


Now NANOROBOTS are believed to be useful against this virus. Size of a nanorobots is 0.1-10 micrometer in diameter .With the development of nanotechnology and nanoelectronics such nanorobots are being developed. This nanorobot will consist a nano biosensor, a nano tube fixed at its tip, and two containers containing high concentration of DNase and RNase enzyme. The function of nano sensor is to identify the presence of a particular compound on the transmembrane (here gp120 & gp41, the unique HIV viral regulatory protein) of the infected cell by means of immunochemical reaction & sending signal. On the receiving end there will be a data convertor which in turn getting +ve signal injects the nanotubes in the nucleus and release DNase and RNase in. These enzymes will cleave the viral genomic DNA & all mRNA produced by it at that time into single nucleotides, as it is not site specific and mutation of HIV genome will have no effect on the efficiency of the enzyme. In this way destroying all the infected cell & HIV genome in the infected body this dieses can be ended.



NANOROBOTICS
Nanorobotics is the technology of creating machines or robots at or close to the microscopic scale of nanometers (10-9 meters). More specifically, nanorobotics refers to the still largely hypothetical nanotechnology engineering discipline of designing and building nanorobots. Nanorobots (nanobots, nanoids or nanites) would be typically devices ranging in size from 0.1-10 micrometers and constructed of nanoscale or molecular components. As no artificial non-biological nanorobots have yet been created, they remain a hypothetical concept.Nanomachines are largely in the research-and-development phase, but some primitive molecular machine has been tested. An example is a sensor having a switch approximately 1.5 nanometers across, capable of counting specific molecules in a chemical sample. The first useful applications of nanomachines, if such are ever built, might be in medical technology, where they might be used to identify cancer cells and destroy them. Another potential application is the detection of toxic chemicals, and the measurement of their concentrations, in the environment.

What is HIV?
HIV stands for Human Immunodeficiency Virus. Like all viruses, HIV cannot grow or reproduce on its own. In order to make new copies of itself it must infect the cells of a living organism. 
HIV belongs to a special class of viruses called retroviruses. Within this class, HIV is placed in the subgroup of lent viruses. 
Outside of a human cell, HIV exists as roughly spherical particles (sometimes called virions). The surface of each particle is studded with lots of little spikes. 
An HIV particle is around 100-150 billionths of a metre in diameter. That's about the same as: 
· 0.1 microns 
· 4 millionths of an inch 
· one twentieth of the length of an E. coli bacterium 
· One seventieth of the diameter of a human CD4+ white blood cell. 
Unlike most bacteria, HIV particles are much too small to be seen through an ordinary microscope. However they can be seen clearly with an electron microscope.

The structure of HIV:
HIV particles surround themselves with a coat of fatty material known as the viral envelope (or membrane). Projecting from this are around 72 little spikes, which are formed from the proteins gp120 and gp41. Just below the viral envelope is a layer called the matrix, which is made from 

the protein p17. 
The proteins gp120 and gp41 together make up the spikes 
that project from HIV particles, while p17 forms the matrix 
and p24 forms the core. 
The viral core (or capsid) is usually bullet-shaped and is made from the protein p24. Inside the core are three enzymes required for HIV replication called reverse transcriptase, integrase and protease. Also held within the core is HIV's genetic material, which consists of two identical strands of RNA. 
Almost all organisms, including most viruses, store their genetic material on long strands of DNA. Retroviruses are the exception because their genes are composed of RNA (Ribonucleic Acid). 
RNA has a very similar structure to DNA. However, small differences between the two molecules mean that HIV's replication process is a bit more complicated than that of most other viruses. 
How many genes does HIV have?
HIV has just nine genes (compared to more than 500 genes in a bacterium, and around 20,000-25,000 in a human). Three of the HIV genes, called gag, pol and env, contain information needed to make structural proteins for new virus particles. The other six genes, known as tat, rev, nef, vif, vpr and vpu, code for proteins that control the ability of HIV to infect a cell, produce new copies of virus, or cause disease. 
At either end of each strand of RNA is a sequence called the long terminal repeat, which helps to control HIV replication.


HIV life cycle:

Entry
HIV can only replicate (make new copies of itself) inside human cells. The process typically begins when a virus particle bumps into a cell that carries on its surface a special protein called CD4. The spikes on the surface of the virus particle stick to the CD4 and allow the viral envelope to fuse with the cell membrane. It is done by two unique HIV envelope protein called gp41 and gp120. Gp120 binds with CD4 and then gp41 comes and facilitates the entry of the virus into the host. After the viral entry the viral protein gp41 and gp120 is found in the cell membrane of the infected cell. The contents of the HIV particle are then released into the cell, leaving the envelope behind. 

Reverse Transcription and Integration
Once inside the cell, the HIV enzyme reverse transcriptase converts the viral RNA into DNA, which is compatible with human genetic material. This DNA is transported to the cell's nucleus, where it is spliced into the human DNA by the HIV enzyme integrase. Once integrated, the HIV DNA is known as provirus. 
Transcription and Translation
HIV provirus may lie dormant within a cell for a long time. But when the cell becomes activated, it treats HIV genes in much the same way as human genes. First it converts them into messenger RNA (using human enzymes). Then the messenger RNA is transported outside the nucleus, and is used as a blueprint for producing new HIV proteins and enzymes. 
Assembly, Budding and Maturation
Among the strands of messenger RNA produced by the cell are complete copies of HIV genetic material. These gather together with newly made HIV proteins and enzymes to form new viral particles, which are then released from the cell. The enzyme protease plays a vital role at this stage of the HIV life cycle by chopping up long strands of protein into smaller pieces, which are used to construct mature viral cores. 
The newly matured HIV particles are ready to infect another cell and begin the replication process all over again. In this way the virus quickly spreads through the human body. And once a person is infected, they can pass HIV on to others in their bodily fluids. 
For the viral infection integration of viral genome in the host genome is necessary and in case of all infected cell the unique viral envelope protein will be present on the cell membrane. This viral protein can be used to identify the infected cell and this job will be done by nanorobot.


THE LATEST DRUG USED AGAINST HIV:

Zidovudine is the latest known drug that is used for treatment of aids. This drug has an affinity to the HIV genome (RNA molecule) and they binds to it before reverse transcriptase starts working and as a result DNA cannot be synthesized. But any time this drug can lose its efficiency as mutation at the codon no. 67,70 and 215 will change the conformation which will result in the reduction of affinity of Zidovudine towards viral genome and as a result RT will start its action and viral genome will be replicated and integrate with host genome.

TREATMENT OF AIDS BY NANOROBOTS:


Zidovudine can be used to resist the HIV but the virus cannot be destroyed. Destruction of viral 
genome is possible by using nanorobots. This type of nanorobots will consists of a nanobiosensor developed by nanoelectronics engineers, a data converter, and a container containing high concentration (say 20 u/microlitre) of DNase and RNase enzyme.

Components of an anti-HIV nanorobot:

Nanobiosensor : The Ab for the Ag gp41 & gp120 will be tagged on its surface. So whenever it will come in contact of an infected cell the Ab will react with that by an immunochemical reaction and will identify this.

Nanochip : It’s a chip which will receive the signal from nanobiosensor and will perform its job.

Nanotube : It’s a tube in nanoscale. On receiving +ve signal the nanotube will be injected into the nucleus of the cell by nanochip.

Nanocontainer : A nanocontainer will contain highly concentrated DNase and RNase enzyme which will be delivered into the infected cell and will cleave the whole genomic DNA into single nucleotides.

Process:
The function of the biosensor is to identify a particular compound. In this case the biosensor will contain a particular antibody. The gp41 and gp120 are two unique HIV envelope protein which is found in the cell membrane of the infected cell. The antigen (gp41 and gp120 protein) and antibody reaction will give the proper signal. In case of infected cell only this reaction will take place as those viral proteins are found in the cell membrane of the infected cell only. Getting the +ve signal the nanorobot will inject its nanotube into the nucleus of the infected cell and release the DNase as well as RNase enzyme into the cell. The DNase enzyme is not sequence specific and as a result it will cleave the whole genomic DNA containing the viral genome into single nucleotides. Once the viral genome loses its sequence it loses its viral effect and after the digestion of the whole genomic DNA the cell undergoes normal programmed cell death called apoptosis. Thus the infected cell of the diseased body can be destroyed to finish off the viral genome in the body.
